The higher-dose arm (650mg) raised free testosterone by 38% and total testosterone by 18%. Both supplemented groups also showed significantly lower cortisol than placebo during the training period.[2] So this trial successfully used the same 325mg daily dose as Combat Tested 3.0.
- Third, a 12-week trial in 92 healthy men and women ages 55-70 using the 650mg dose alongside a structured at-home exercise program, demonstrated that these effects extend into older populations. That group achieved a 4.2x improvement in lower body strength and a 4.5x improvement in grip strength over placebo. Testosterone increased significantly in the male subjects (p < 0.009).[3]
- The most directly relevant data for Combat Tested's target user comes from a 2025 clinical trial published as an abstract in Current Developments in Nutrition. This 8-week, placebo-controlled study enrolled 60 healthy, recreationally active men ages 40-65 -- the demographic this product is built for -- at the exact 325mg daily dose. Total testosterone improved significantly versus placebo by week 2, and free testosterone by week 4, with both continuing to rise through week 8.[4]

Mechanisms and Formula Synergies
TestFACTOR's effects operate through multiple parallel pathways. Mangiferin from the mango bark extract drives potent antioxidant activity and supports mitochondrial biogenesis, making cellular energy production more efficient. Ginger Root Extract (Zingiber officinale) - 500mg

A 4-week trial in 63 moderately stressed adults found tongkat ali supplementation raised testosterone by 37% while reducing cortisol by 16%, with significant improvements in tension, anger, and confusion scores versus placebo.[15] A 12-week RCT in 109 men ages 30 to 55 using 300mg daily showed significant improvements in erectile function, libido, sperm motility, and physical functioning.[16] A 2022 meta-analysis of five RCTs confirmed a significant overall increase in serum total testosterone (SMD = 1.352).[17]

Shilajit Extract (Asphaltum) - 250mg
Shilajit is a mineral-rich Himalayan exudate whose key bioactives, fulvic acid and dibenzo-α-pyrones (DBPs), support both testosterone levels and mitochondrial energy production.
A 90-day, double-blind, placebo-controlled trial in 75 healthy men aged 45-55 found purified shilajit at 500mg daily significantly raised total testosterone by 20.45%, free testosterone by 19.14%, and the precursor hormone DHEAS by 31.35% versus placebo, with LH maintained throughout.[18] A separate 8-week trial in 63 active men found that 500mg/day reduced fatigue-induced strength decline to 8.9% versus 16.0% for placebo, while also lowering serum hydroxyproline, an indirect marker of collagen breakdown.[19]
The mechanism behind both effects traces back to fulvic acid's role as a cellular transporter: it improves mineral uptake and supports coenzyme Q10 (CoQ10) availability within the mitochondria, which the electron transport chain depends on for efficient ATP synthesis. Preclinical work using the ingredient showed significantly greater post-exercise muscle ATP concentrations and a meaningfully higher adenylate energy charge versus unsupplemented controls, supporting the idea that enhanced mitochondrial efficiency drives the fatigue-resistance outcomes seen in human trials.[19]
The 250mg dose here falls below the 500mg threshold where both human outcomes reached significance, so its role in Combat Tested 3.0 is reinforcing rather than primary. That said, shilajit's mitochondrial ATP support directly complements TestFACTOR's central mechanism of mitochondrial biogenesis, adding a second input to the same energy and testosterone pathway that anchors this stack.

3,3'-Diindolylmethane (DIM) - 100mg
3,3'-Diindolylmethane (DIM) is the bioactive metabolite formed when indole-3-carbinol (I3C) (a compound found in cruciferous vegetables like broccoli and cabbage) undergoes acid condensation in the stomach.[20] Its job in this formula is estrogen management: as the other ingredients push testosterone higher, increased aromatase activity can convert more of that testosterone to estradiol, shifting the hormonal balance in the wrong direction. DIM helps counteract this by influencing how estrogen gets metabolized downstream.
A shift towards better estrogen
Specifically, DIM favors the 2-hydroxy pathway, steering metabolism toward 2-hydroxyestrone (2-OHE1), a comparatively weak estrogen metabolite, and away from 16α-hydroxyestrone (16α-OHE1), which binds estrogen receptors more aggressively. This favorable shift in the 2-OHE1:16α-OHE1 ratio has been confirmed in human trials.[21][22]

Vitamin D3 (cholecalciferol) fills a micronutrient gap that quietly undermines hormonal health in most men's diets, especially those who aren't out in the sun a bunch. The vitamin D receptor (VDR) and key vitamin D-metabolizing enzymes are expressed throughout the human male reproductive tract, including the Leydig cells where testosterone synthesis occurs.[25] That gives 25-hydroxyvitamin D [25(OH)D] a direct upstream role in androgen production.
Cross-sectional data from 2,299 men found vitamin D status tracks with testosterone and free androgen index, with both following the same seasonal arc, peaking in August and bottoming out in March.[26] In a 12-month RCT, men who started out deficient and supplemented with 3,332 IU daily saw significant increases in total testosterone (~25%), bioactive testosterone, and free testosterone compared to placebo.[27] A 2019 meta-analysis of eight RCTs found no overall significant effect across the broader study population, which suggests baseline deficiency status determines how much room there is for improvement.[28]
At 40mcg (1,600 IU), this dose covers daily maintenance needs for most men. The boron in this formula extends its working window by inhibiting its breakdown, bringing the synergy discussed in the section above.

Regular exercise increases urinary zinc losses and redistributes zinc away from circulation, raising functional depletion risk in athletes and active men.[30] Research in sedentary men showed that four weeks of zinc supplementation maintained both resting and post-exercise testosterone levels that otherwise declined significantly with fatigue.[31] The picolinate form pairs zinc with picolinic acid, one of the body's natural mineral-transport molecules, for improved absorption. At 15mg, it's the right dose to maintain the hormonal foundation that TestFACTOR, Tongkat Ali, and Shilajit are building on top of.
