MuscleTech Metabolic Peptide 320: Stim-Free Metabolic Activation with TheoLim® & DNF-10™

The metabolic support category has a stimulant problem. Most fat burners lean on caffeine-heavy stacks to drive output, which works for some users but leaves others (those sensitive to stimulants or who already take a pre-workout) with limited options. MuscleTech Metabolic Peptide 320 takes a different approach, targeting biological signaling rather than stimulation to support resting metabolism.

MuscleTech Metabolic Peptide 320: Stim-Free Metabolic Activation with TheoLim® & DNF-10™

MuscleTech Metabolic Peptide 320 skips the stimulants entirely. TheoLim® burns 3.2x more calories at rest. DNF-10™ targets hunger signals from the inside out. Two ingredients, two distinct jobs, one clean daily capsule.

The formula pairs two research-backed ingredients. TheoLim® is a patented blend of lime peel extract (Citrus aurantifolia) and cocoa seed extract (Theobroma cacao) that supports natural GLP-1 release and adiponectin production to activate AMPK, the body's primary metabolic switch. PricePlow readers will recognize TheoLim from our coverage of MuscleTech EFF'N Cut!, where it drove the same headline claim in an effervescent format. Here, it's paired with DNF-10™, a bioactive peptide from hydrolyzed brewer's yeast (Saccharomyces cerevisiae) that targets hunger regulation and appetite control.

Peptides First, Thermogenics Second

The two-ingredient design reflects the logic behind MuscleTech's Peptide Series: use targeted biological tools instead of blunt stimulation. In the MP320 system, DNF-10 acts as the appetite-regulation layer, supporting hunger control and gut-brain metabolic signaling, while TheoLim delivers the thermogenic output. Clinical research on TheoLim shows test subjects burning 3.2x more calories at rest versus placebo after a single dose, with a separate 16-week trial demonstrating even larger gains over time.

One clarification worth noting upfront: this product contains no GLP-1 and is not a GLP-1 agonist. It supports the body's own natural GLP-1 production within a normal physiological range, which is an important distinction given current regulatory attention on GLP-1-adjacent claims. MuscleTech Metabolic Peptide 320 Ingredients

Each 2-capsule serving provides the following:

MuscleTech Metabolic Peptide 320 Ingredients

  • TheoLim® (Lime Peel Extract and Cocoa Seed Extract) - 450mg

TheoLim® is a patented botanical blend combining aqueous-ethanol extract of Citrus aurantifolia fruit rind and water extract of Theobroma cacao seed at a 2:1 ratio. The composition is standardized to at least 5.0% citric acid, 0.5% theobromine, and 0.3% hesperidin by HPLC, and is produced at a cGMP-certified facility.[1]

    How It Works

TheoLim targets several metabolic pathways simultaneously rather than relying on stimulant-driven thermogenesis. The blend supports natural GLP-1 secretion in enteroendocrine cells through activation of bitter taste receptors (hTAS2Rs) and downstream phospholipase C signaling.[2] Elevated GLP-1 upregulates GLP-1 receptor expression in the liver, while rising adiponectin levels activate AMPK, the primary cellular energy-sensing enzyme.

Downstream, AMPK signaling increases expression of UCP-1 (uncoupling protein-1) and PGC-1α in adipose tissue, promoting heat dissipation and fat oxidation rather than ATP production, the core of its thermogenic effect.[3]

    Clinical Evidence

In a randomized, double-blind, placebo-controlled crossover trial of 60 overweight adults (BMI 25–29.9 kg/m²), a single 450mg dose increased resting metabolic rate (RMR) by 10.19%, 19.44%, and 24.07% at 60, 120, and 180 minutes post-dose versus pre-dose -- with significant between-group differences at 120 and 180 minutes versus placebo. Fat oxidation increased by 17.68% and 20.91% at those same time points. Importantly, blood pressure and heart rate remained comparable between groups, confirming a stimulant-free thermogenic profile.[1]

    MuscleTech Metabolic Peptide 320

A subsequent 16-week multicenter RCT in 120 overweight adults (450mg daily) found significantly greater reductions in body weight (4.25 ± 1.35 kg vs. 0.96 ± 1.18 kg for placebo), total body fat (4.28 ± 1.56 kg vs. 0.85 ± 1.06 kg), and waist circumference (4.32 cm vs. 2.10 cm) versus placebo, while lean body mass was preserved. Serum GLP-1 increased 33.05% and adiponectin rose 51.90% from baseline at 16 weeks, while ghrelin declined significantly.[4]

    Dosing and Synergy in MP320

The clinical-validated dose is 450mg taken in a fasted state before the first meal, matching exactly what MP320 provides in a two-capsule serving. In this formula, TheoLim's GLP-1 support complements DNF-10's appetite-regulation mechanism, targeting two distinct but interconnected signals: resting energy output and hunger control, both relevant to long-term body composition management.

  • DNF-10™ Yeast Hydrolysate Peptide - 250mg

DNF-10™ is a bioactive peptide complex derived from hydrolyzed brewer's yeast (Saccharomyces cerevisiae). It holds FDA GRAS status (GRN 1033),[5] and its appetite-regulating mechanism works through central nervous system and gut-brain signaling rather than adrenergic stimulation.

    How It Works

DNF-10 modulates appetite-regulating neuropeptides in the hypothalamus. In rat studies, yeast hydrolysate at this size (below 10 kDa) significantly reduced neuropeptide Y (NPY) expression in the paraventricular nucleus and lateral hypothalamic area (a potent orexigenic signal that drives feeding behavior) while simultaneously increasing tryptophan hydroxylase (TPH) activity in the dorsal raphe, the rate-limiting enzyme in serotonin synthesis and an opposing appetite-suppressing signal.[6]

MuscleTech EFF'N Cut! Effervescent Fat Burner with 3.2x Calorie-Burning Power

MuscleTech EFF'N Cut! brings clinical thermogenics to an effervescent format. TheoLim increased metabolic rate 3.2x above baseline in studies, while the FIZZR beadlet system makes daily fat burning something you'll actually look forward to.

DNF-10 also contains cyclo-His-Pro (CHP), a bioactive dipeptide associated with leptin-like activity and dopaminergic regulation in the CNS. Additional mechanisms include reduced ghrelin secretion and attenuation of hepatic lipogenic enzyme activity.[7]

    Clinical Evidence

In a 10-week randomized, placebo-controlled trial (n=54 obese adults), 1g/day yeast hydrolysate produced significantly greater body weight loss (-2.60 kg vs. -0.83 kg for placebo) and CT-confirmed reductions in total abdominal fat area (-17.34 cm² vs. -7.06 cm²; P < 0.01), while lean body mass was preserved.[7] Reduced calorie intake drove the effect, diverging significantly from placebo by week 6 with no changes to physical activity.

At a lower 500mg/day dose, an 8-week RCT in obese women produced significant reductions in body weight, fat mass, and fat mass ratio versus placebo, with subjects also reporting reduced food cravings and a meaningful drop in sweet preference.[8] A 2025 meta-analysis pooling clinical trials found mean reductions of -3.08 kg body weight (P < 0.001), -0.87 kg/m² BMI, and -1.93 kg fat mass, with a favorable safety profile across all trials.[9]

    Dosing and Synergy in MP320

MP320 delivers 250mg of DNF-10 per serving, with an optional second serving bringing the daily total to 500mg, matching the lower-dose clinical threshold where meaningful outcomes were still observed. In this formula, DNF-10's intake-side signaling complements TheoLim's thermogenic output: TheoLim addresses resting energy expenditure through GLP-1 and AMPK pathways, while DNF-10 targets calorie intake through NPY suppression, serotonin upregulation, and ghrelin modulation. Two distinct levers on the same body composition goal.

MuscleTech Metabolic Peptide 320 takes a tight, purposeful approach to metabolic support: TheoLim® drives the thermogenic output, and DNF-10™ addresses the hunger-regulation side that most thermogenics skip entirely. Two ingredients, two distinct jobs, both backed by human clinical research, in a once-daily capsule format that keeps stimulants out of the equation.

That stimulant-free profile also makes it more stackable than a typical fat burner. It doesn't conflict with a pre-workout, doesn't require cycling around other stimulant products, and fits naturally alongside other daily-use peptide supplements. If you're building out a full peptide stack, Test Peptide T10 makes a strong starting point alongside MP320 for daily performance and body composition support.
